Fugu is a very useful model for gene discovery, and for annotating the human and other vertebrate genomes. Apart from annotating coding sequences, Fugu is also useful for identifying and characterizing gene regulatory elements. Since the intergenic and intronic regions are highly compressed in the Fugu, it is easy to identify conserved non-coding sequences in the Fugu and to assay them for function. Transgenic studies in rodents and frogs, as well as transfection into mammalian cell lines, have shown that mammalian and amphibian trans factors are able to interact with Fugu cis elements to mediate cell - and tissue-specific expression, and to mediate appropriate response to physiological stimuli. Since a Fugu cosmid clone typically contains 6 to 8 genes and a BAC clone contains up to 15 genes, Fugu genomic clones are ideally suited for dissecting gene regulatory regions and locus control elements. Which one day help us get closer to understanding Cancer if we can decipher the coding.
